LB-102 Overview

A purpose-built approach to multi-dimensional treatment

LB-102

Purposeful Innovation

An investigational new chemical entity built on one of the most well studied foundations in psychiatry.

receptor

Unique Receptor Pharmacology

Selective D2/D3/5-HT7 pharmacology intended to address symptom domains that matter across psychosis and mood disorders.

Designed for Real-World Use

Evaluating LB-102’s potential to optimize efficacy, tolerability, and ease of use.

LB-102 demonstrated statistically significant benefit versus placebo. An effect was observed as early as week 1, with antipsychotic efficacy sustained through the endpoint of the trial. In this trial, LB-102 also demonstrated a favorable safety and tolerability profile, characterized by low rates of extrapyramidal symptoms (including akathisia), minimal sedation and few gastrointestinal side effects. Effects on negative symptoms and cognitive performance were also observed. These data support further investigation in a larger number of patients with schizophrenia and in trials of LB-102 in mood disorders.

LB-102 & Amisulpride

Heritage in a well studied foundation

Amisulpride helped establish the therapeutic potential of selective dopamine and serotonin receptor inhibition for psychosis and mood disorders. It is, however, limited by poor blood brain barrier penetration. As a result, amisulpride requires high doses that carry greater systemic exposure and must be taken twice daily.

LB-102 is a new chemical entity that was purposefully designed to cross the blood brain barrier, with the goal of enabling efficacy at lower systemic drug levels with simple once daily dosing while preserving selective inhibition of D2/D3 and 5-HT7.

Potential innovations of LB-102 that are being assessed in ongoing clinical trials:

  • Improved potency, lowering required dose and systemic exposure
  • Once-daily dosing
  • Improved tolerability

LB-102 Mechanism

Potential mechanism of action in brain disorders

LB-102 selectively targets three receptors involved in neuropsychiatric function:

D2 Receptor

D2

A dopamine receptor associated with psychosis (positive symptoms) and mania

D3

A dopamine receptor associated with pathways related to depression, cognition, anhedonia, and negative symptoms

5-HT7

5-HT7

A serotonin receptor associated with pathways involved in depression, cognition, anhedonia, and negative symptoms

LB-102 may have a binding profile that modulates dopamine activity differently depending on dose. This potential is the premise for evaluating different doses across multiple brain disorders.
